Jove is “a poetical equivalent of Jupiter, name of the highest deity of the ancient Romans” ("Jove, n."). In this scenario, “by Jove” is meant to signify that the speaker is shocked or surprised. It is a variation of saying “my god” without actually invocating god himself.
